KUALA LUMPUR (THE STAR/ASIA NEWS NETWORK) - She was a successful swimmer turned fitness buff but a tragedy robbed Ms Nur Hidayah Tawiei, 25, of a promising future.

Ms Nur Hidayah fell gravely ill and as her condition worsened, she was kept alive for a year before succumbing to renal failure in 2014.

Last Saturday (Nov 11), her family held an event to raise funds for the treatment of three children suffering with the same condition.

They are among 1,600 children in Malaysia who will receive dialysis treatment for at least five years, as a result of poor kidney function.

The treatment costs RM400,000 per session, and each child needs at least four sessions a week.

Ms Nur Hidayah's parents, Tuan Abu Bakar and Siti Zulaikha Tawiei, are still struggling to pay off the hefty bills, and are appealing to the public for help.

"The treatment is very expensive and life-saving for our kids, but it can be cured with proper treatment," her mother, Tuan Abu Bakar, said.

"We're looking for help from the public to raise funds. If we raise more money, we can give our kids proper treatment," he added.

Nur Hidayah died when she was just 21, after contracting a kidney infection which made her kidneys fail.

Her mother said that since her death, the family's two elder children, Jumuah and Zulaikha, are suffering from a poorly functioning liver, and the youngest, Nur Azah, has chronic cystitis, which has also affected her kidneys.

"We are grateful to Aishah Dian for nominating us for this year's Kaspers, as she donated RM1,000 (S$250) for the fight against renal failure.

"But we need your help to continue this struggle and raise funds for the treatment of Nur Hidayah's three kids," Tuan Abu Bakar added.

The mammalian striatum generates two functionally and anatomically distinct neural networks. The output circuits of these systems are distributed in the basal ganglia and mediate crucial aspects of motor control, including initiation, inhibitory control, and timing of movement. The basal ganglia are thus crucial for a vast array of functions and diseases affecting motor behavior. Major progress has been achieved in understanding the essential role of dopamine in regulating output circuitry in the striatum. With the advent of gene targeting and transgenic approaches, mice have emerged as a powerful animal model for studying the function of these circuits. However, mice are not a good model for studying the circuit functions in primates, such as humans. Moreover, compared with other murine brain regions, such as hippocampus, cortex, and cerebellum, the mouse striatum is in an extremely early stage of development and its gross anatomical organization is almost totally unknown. Here we propose to take a multi-disciplinary approach to understand the molecular, cellular, and circuit mechanisms that control output circuits in the striatum. Using a combination of genetic approaches with transgenic mice, in vitro slice electrophysiology, and in vivo microdialysis, we will examine the following hypotheses: 1) Presynaptic GABA transporters control the GABAergic output circuit. We will analyze the molecular properties of these transporters and generate genetic mouse models to test this hypothesis. 2) Dopaminergic neurons are crucial for the GABAergic output circuit. We will determine which subtypes of dopamine neurons are involved in the GABAergic output circuit and manipulate the activity of these neurons in mice. 3) The glutamatergic output circuit is modulated by striatal-forebrain-cortical loops. We will examine the anatomical organization of this circuit and determine which of the different types of striatal output neurons projects to the cerebral cortex, and which of the different cortical regions send axonal projections to the striatum. 4) The GABAergic and glutamatergic output circuits influence the activity of the dopamine neurons, and feedback to the striatal dopamine input circuits. We will examine the relationship between the GABA and glutamatergic output circuits, and the dopamine neurons in the striatum using optogenetic techniques.
