In the present study, we provide a novel approach to study synaptic plasticity in the hippocampus. This consists in the use of small extracellular electrodes (2 μm tip diameter) positioned in CA1 stratum oriens to electrically stimulate Schaffer collaterals and monitor the frequency of EPSCs recorded in CA1 pyramidal cells. This approach allowed us to obtain reliable EPSCs frequency in a constant range of physiological conditions in both acute and long-term chronic slices, and it is well suited to record the effect of theta-burst stimulation (TBS) on plasticity. Here we show that this novel approach for monitoring synaptic plasticity is a valid and reliable tool to investigate the effect of TBS on synaptic transmission and plasticity. Furthermore, the study of synaptic plasticity is not altered by using this approach instead of the conventional whole-cell patch clamp.[Endobronchial recurrence
